Disc Medicine eyes two pivotal programmes by 2027 after busy Q2

The Watertown biotech reported $718m in cash runway to 2029 and flagged a Q4 Phase 3 readout for bitopertin in EPP as its nearest catalysts.

Disc Medicine has reported its second-quarter 2026 results alongside a substantive pipeline update, with the NASDAQ-listed hematology specialist pointing to a Q4 2026 Phase 3 data readout for its lead asset bitopertin and the potential to advance two further candidates into pivotal development in 2027.

The company closed the quarter with $717.7 million in cash, cash equivalents, and marketable securities, a position it says will fund operations into 2029. Net loss for the three months to 30 June was $59.5 million, up from $55.2 million in the same period last year, driven by higher headcount and continued clinical investment across its three-programme pipeline.

Bitopertin and the CRL pathway

Bitopertin, a GlyT1 inhibitor that modulates heme synthesis, remains the company's most advanced asset. Disc completed a Type A meeting with the FDA during the quarter, aligning with the agency that a successful Phase 3 APOLLO study could form the basis for a response to the complete response letter (CRL) previously received for the drug in erythropoietic protoporphyria (EPP). APOLLO enrolled 183 patients ahead of schedule and closed enrolment in March 2026; topline data are expected in Q4 2026, with a CRL response submission and an FDA decision targeted for mid-2027.

Disc also launched an Expanded Access Programme for bitopertin in the US and select international markets, providing eligible EPP and XLP patients access to the drug ahead of any regulatory approval. Updated data from the HELIOS open-label extension trial, presented at the European Hematology Association (EHA) annual meeting, showed sustained reductions in protoporphyrin IX, meaningful improvements in light tolerance, and a favourable longer-term safety profile.

Chief executive John Quisel said the company was positioned to "advance two more programs into pivotal-stage development in 2027," referencing selcodebart and DISC-3405 alongside the expected bitopertin regulatory decision.

Pipeline breadth across hematology

Selcodebart (DISC-0974), an anti-hemojuvelin antibody that suppresses hepcidin, continued to generate data in anemia of myelofibrosis. Results shared at both ASCO and EHA showed a major anemia response rate of 56% and an overall anemia response rate of 72% across evaluable patients in the RALLY-MF Phase 2 trial, with consistent responses regardless of transfusion status or background JAK inhibitor use. An end-of-Phase 2 meeting with the FDA is expected by year-end, and the candidate received EU Orphan Drug Designation for myelofibrosis during the period. A Phase 2 study in inflammatory bowel disease is also progressing, with initial data anticipated in 2027.

DISC-3405, an anti-TMPRSS6 antibody that induces hepcidin, completed enrolment in the RESTORE-PV Phase 2 study in polycythemia vera ahead of schedule, with initial data now expected in Q3 2026. A Phase 1b study in sickle cell disease is advancing in parallel, with initial data expected in Q4 2026.

Market context

Disc operates in a hematology space that has attracted sustained investor interest, particularly around rare red-blood-cell disorders where approved treatment options remain limited. EPP has no FDA-approved therapy following earlier setbacks in the category, making bitopertin's registrational path closely watched. The myelofibrosis anemia segment is also competitive, with several companies pursuing hepcidin-pathway and erythropoiesis-stimulating approaches; the RALLY-MF response rate data will need to be contextualised against durability and safety endpoints when fuller datasets emerge later this year. With three programmes generating near-term binary readouts and a cash position that removes financing risk through 2029, Disc enters the second half of 2026 in an operationally strong position, though the outcome of the APOLLO trial remains the single most consequential event on its near-term horizon.
