IMUNON signs NCI CRADA to expand IMNN-001 into new solid tumours
IMUNON has entered a Cooperative Research and Development Agreement (CRADA) with the National Cancer Institute, giving the Lawrenceville, New Jersey biotech access to NCI clinical infrastructure to test its lead candidate, IMNN-001, across new solid tumour types and combination regimens.
Under the agreement, NCI's Cancer Therapy Evaluation Program (CTEP) assumes regulatory sponsorship and will run additional clinical and non-clinical studies through the NCI Experimental Therapeutics Programme. IMUNON retains ownership of its background intellectual property and any new IP generated solely by the company during the collaboration, including rights to use all resulting data for its own development programmes. IMNN-001 is described as the first gene-based immunotherapy selected by NCI/CTEP for partnered clinical development under a CRADA.
The deal
The collaboration builds on clinical data from IMUNON's ongoing ovarian cancer programme. In the completed Phase 2 OVATION 2 study, IMNN-001 added to neoadjuvant and adjuvant chemotherapy was associated with a 14.7-month improvement in median overall survival compared with chemotherapy alone (45.1 versus 30.4 months) in 112 patients with newly diagnosed advanced ovarian cancer. The candidate is now in the pivotal Phase 3 OVATION 3 trial in the same frontline setting.
IMNN-001 uses IMUNON's TheraPlas platform: a DNA plasmid vector encoding IL-12, encased in a nanoparticle delivery system designed to transfect local cells and drive persistent, localised secretion of the cytokine. IL-12 stimulates T-lymphocyte and natural killer cell proliferation, representing a mechanistic rationale the company believes extends well beyond ovarian cancer.
Chief medical officer Douglas Faller said: "The mechanistic rationale for IL-12 as a multipotent immune activator extends well beyond ovarian cancer, and this collaboration gives us a capital-efficient way to initiate additional clinical trials for IMNN-001 across diverse solid tumour types and indications."
Market context
NCI CRADAs are a recognised route for smaller biotechs to access the National Clinical Trials Network without bearing the full operational and regulatory cost of running independent studies. For a Nasdaq-listed clinical-stage company like IMUNON, the arrangement is particularly meaningful: the burden of funding new-indication trials shifts substantially to NCI, preserving cash for the pivotal OVATION 3 programme. CEO Stacy Lindborg indicated the company's internal team will remain focused on that Phase 3 study.
The IL-12 immunotherapy space has a long and at times difficult history. Earlier systemic IL-12 approaches were abandoned due to toxicity; local delivery strategies, such as those employed by TheraPlas, have renewed interest in the cytokine by aiming to concentrate activity at the tumour site while limiting systemic exposure. Several academic groups and a number of biotechs are pursuing localised cytokine delivery strategies in solid tumours, making the competitive environment active even if no direct approved comparator exists.
For investors, the key near-term readouts remain tied to OVATION 3. The CRADA extends the pipeline's optionality without requiring fresh capital, but indication selection and combination partners under the NCI programme have not yet been disclosed. Clarity on which tumour types CTEP will prioritise, and any early efficacy signals from those studies, will be the metrics to watch as this collaboration matures.
